Product Background

Hepatitis B virus (HBV) is one of the primary pathogens responsible for acute and chronic hepatitis in humans. HBV exhibits strong species specificity, naturally infecting only primates (humans and chimpanzees). However, due to ethical and economic constraints, research into HBV infection biology and immunology using chimpanzees is severely limited. While the application of the tree shrew model has led to significant progress in understanding the mechanisms of chronic HBV infection, tree shrews have a low infection rate and only experience transient infections. Therefore, the development of simple, cost-effective mouse models is a key trend in HBV research.

Construction Principle

By intravenously injecting AAV-HBV (adeno-associated virus carrying the HBV genome), the HBV viral genome can be delivered to mouse liver cells, thereby simulating HBV infection. This method yields a genetically uniform mouse model that is widely used in HBV drug research. Currently, the hepatitis B virus mouse model stands as one of the most successful applications of AAV in constructing animal models for liver diseases. Hepatitis B is an infectious disease primarily characterized by inflammatory lesions in the liver, caused by the Hepatitis B virus (HBV), and can lead to multi-organ dysfunction. By utilizing recombinant AAV serotype 8 carrying 1.3 copies of the HBV genome (rAAV8-1.3HBV), a persistently infected HBV mouse model can be established. This AAV-mediated modeling approach not only boasts high efficiency but also maintains HBV infection for an extended period, offering a novel animal model for research into HBV infection and pathogenic mechanisms, as well as for the screening of antiviral drugs and vaccines.
