Lentivirus packaging service

Lentiviral vectors (LVs) are a viral vector system engineered from the human immunodeficiency virus (HIV-1). They can very efficiently and stably integrate exogenous genes into mammalian cells and are capable of infecting both dividing and non-dividing cells. Due to their broad tropism and flexible selection for target genes, LVs are now widely used in research fields such as stable transgene overexpression, sustained gene silencing, gene editing, animal immunization, imaging tracing, and induced pluripotent stem cell modification.

Note: The third-generation packaging system exhibits higher specificity, exclusively supporting third-generation transfer vectors. In contrast, the second-generation system offers broader compatibility, accommodating both second and third-generation transfer vectors. Key modifications in third-generation transfer vectors involve a critical alteration in the 5' LTR region, where the U3 element is replaced with an exogenous strong promoter like CMV or RSV. This structural optimization frees the transcription of the viral genome from dependence on the Tat protein. Consequently, the core plasmid components of third-generation packaging systems no longer include the Tat protein coding sequence. Conversely, second-generation transfer vectors retain the complete wild-type LTR structure (including U3-R-U5 elements), and their transcriptional activation remains dependent on the trans-regulatory action of the Tat protein.
