Adeno-Associated Virus (AAV) is a non-enveloped virus belonging to the genus Dependovirus within the Parvoviridae family. It has a particle diameter of 18-26 nm and a genome size of 4.7 kb. Various serotypes exhibit distinct host specificities and characteristics. Recombinant AAV vectors (rAAV) have emerged as a leading platform for in vivo gene therapy delivery due to their low pathogenicity, minimal immunogenicity, stable long-term expression, and broad tissue tropism.
Traditional purification of AAV viral particles is typically performed using iodixanol density gradient centrifugation, a labor-intensive and time-consuming process with relatively low recovery rates. The AAV Magnetic Bead Purification Kit streamlines this procedure, enabling virus purification within approximately one hour. This method is suitable for a broad range of serotypes, offering simplicity, speed, and high purification efficiency. The purified viral particles can be directly utilized in downstream applications.

Experimental Validation
1、Purity Assessment
SDS-PAGE analysis confirmed the presence of AAV capsid proteins VP1, VP2, and VP3 as dominant electrophoretic bands.
2、Infectivity Evaluation
Purified AAV9-GFP virus was used to infect 293T cells following gradient dilution, demonstrating that viral activity remained unaffected.
