This product is a chimeric virus constructed using an adeno-associated virus (AAV) backbone, where different serotype capsid proteins are substituted based on targeting requirements. This pseudovirus structure possesses single-round infectivity, allowing it to mimic the neutralization process of wild-type viruses by antibodies after cellular entry. Additionally, the pseudovirus contains a luciferase reporter gene nucleic acid sequence, which facilitates the subsequent organization and analysis of neutralization experiment data. This product offers good safety and is easy to operate.

Detection principle

Neutralizing antibodies, generated either through pathogen infection or vaccination, can specifically recognize antigenic sites on the viral surface, thereby blocking viral entry into cells and preventing replication. This protects the host from harm and confers genuine antiviral activity. This AAV pseudovirus contains the viral neutralizing antigen and possesses the ability to infect cells for a single round. When samples with neutralizing activity interact with the pseudovirus, a neutralization reaction occurs, reducing the AAV pseudovirus's infectivity. This leads to a decrease in the expression of the reporter gene carried, Luciferase. By reading the luciferase expression level in each well using a multi-functional microplate reader, and comparing the values from the test sample wells to those of the pseudovirus control group, one can determine if the sample possesses neutralizing activity and calculate its effective concentration. The neutralizing antibody titer is defined as the reciprocal of the antibody dilution that inhibits 50% of pseudovirus infection.
